Making peptides · Part 7: Freeze-drying
Three stages, and what can go wrong
Lesson 4.7.2 · 6 min read · Advanced plan
What you'll learn
- Freeze-drying has three stages: freezing, primary drying (sublimation under vacuum) and secondary drying (removing bound water).
- The product must stay below its critical temperature during primary drying, or it can collapse or melt back.
- Residual moisture limits, leak testing and 100 percent visual inspection of vials are part of a controlled process.
- For hygroscopic peptide powders, water content is part of stability studies.
